re-engineer cells in Real time with

the cybercyte system

The powerful Cybercyte System is a user-friendly, plug and play tool which combines in silico synthetic cells and in vitro patch-clamped live cells, in real time, to maximize the value of your electrophysiological experiments with electronic “transfection” in seconds, not weeks!

The Cybercyte can be configured to connect up to eight in vivo voltage clamped cells with in silico synthetic channels, synapses, and cells. A few examples are given below. We will work with you to configure the system you need for your particular experimental needs.

Validated Cybercyte Ion Channel Models

Insert Synthetic in silico Channels into Whole Cell Patch Clamp Assays

Synthetic in silico heterologously-expressed channels can be electronically transfected into in vitro patch clamped live whole cells. The Cybercyte has over 00 pre-installed validated ion channel models to choose from, or you can add your own based on your own data or on published data. You can use the CyberSolver model building software to create your own Markov Models from your own experimental data or from published data. Cytoybernetics can also build and validate models for you. Using the Cybercyte in this mode is like transfecting a channel into a whole cell, but in seconds rather than weeks. The in vivo current amplitude in the whole cell can be scaled instantly, to reflect changes in expresion.

Insert in vitro Patch Clamped Channels into Synthetic in silico Whole Cells

The Cybercyte goes beyond a simple dynamic clamp system. The Cybercyte can run synthetic in silico whole cell models, and incorporate real time currents from in vitro patch clamp experiments on heterologously expressed cells to create and in silico in vitro hybrid whole cell. The patch clamped cell is instantly electronically “transfected” into the synthetic cell. The Cybercyte has several synthetic in silico whole cell models available, (including synapses) or you can create your own from the library of individual ion channels in the Cybercyte channel library.

Insert Current from Modified in vitro Patch Clamped Channels into Synthetic in silico Cells in Real Time

When using the Cybercyte in Synthetic Cell mode, the in vitro experimental channel can be exposed to drugs, changes in temperature, or any other interventions, and these changes can be examined in the context of the whole cell assay.

Adding Ik1 to cardiac ipscs

Using the Cybercyte to electronically express Ik1 into iPSC cardiac myocytes lowers the resting potential and puts the channels in a more physiologically realistic starting point for the action potential. It also removes automaticity, making analysis easier and quicker as fewer cells are needed.

application of a gating-altering toxin to an in vivo ghannel changes AP firing in the synthetic in silico neuronal cell

A synthetic in silico neuronal cell with a synaptic current had an in vivo patch clamped Nav1.2 electronically expressed in to it, producing stochastic APs. The gating of the in vivo channel was modified by a toxin, which resulted in changes in the firing frequency and action potential morphology of the in silico synthetic cell.
